Direct amplification of nodD from community DNA reveals the genetic diversity of Rhizobium leguminosarum in soil
Auteurs: Adolphe Zézé, L.A. Mutch, J. Peter W. Young
Date publication: 2001-06-01
Journal: Wiley
Sequences of nodD, a gene found only in rhizobia, were amplified from total community DNA isolated from a pasture soil. The polymerase chain reaction (PCR) primers used, Y5 and Y6, match nodD from Rhizobium leguminosarum biovar trifolii, R. leguminosarum biovar viciae and Sinorhizobium meliloti. The PCR product was cloned and yielded 68 clones that were identified by restriction pattern as derived from biovar trifolii [11 restriction fragment length polymorphism (RFLP) types] and 15 clones identified as viciae (seven RFLP types). These identifications were confirmed by sequencing. There were n...

Drivers of tree presence and densities: the case of cocoa agroforestry systems in the Soubre region of Republic of Côte d’Ivoire
Auteurs: Amos Gyau, Kaitlyn Smoot, Lucien Diby, Christophe Kouamé
Date publication: 2014-10-06
Journal: Springer Science and Business Media LLC
Cocoa yields in Côte d'Ivoire are low and falling each year, in part as a result of full-sun cropping systems. Thus, interest is now high in establishing sustainable cocoa agroforests through the re-introduction of shade trees. This article uses data collected from a sample of 400 cocoa farmers in the Soubré region of Côte d'Ivoire to rank the top alternative tree species of interest to farmers and to analyze the determinants of their presence and density in cocoa farms. Results show that the most significant determinants are: social network effects, ethnic group, and geographic zone. Also, po...

DNA cloning and screening of a partial genomic library from an arbuscular mycorrhizal fungus, Scutellospora castanea
Auteurs: Adolphe Zézé, H. Dulieu, Vivienne Gianinazzi-Pearson
Date publication: 1994-08-01
Journal: Springer Science and Business Media LLC
AbstractA technique has been developed to efficiently extract purified, restrictable genomic DNA from spores of different arbuscular mycorrhizal fungi in order to begin detailed investigations of the genome of the Glomales. The protocol yielded variable amounts of DNA depending on the fungal species; for Scutellospora castanea and Gigaspora rosea it reached values of 1.5–2 ng/spore. EcoRI digests of DNA from S. castanea were cloned into pUC18 and about 1000 recombinant DNA clones were obtained. Of those screened, 50 contained inserts of 500–7000 bp. Selected inserts detected DNA sequences from...

Studying both soil carbon (C) and nitrogen (N) storages in different climate zones and their relationship with climatic factors is of great significance for understanding soil fertility and predicting global climate change. Climate influences soil minerals, which are important for soil organic carbon (SOC) and N retention. However, there are few studies on SOC and soil total nitrogen (STN) storage in different climatic zones, and of the effects of soil oxidation minerals on SOC and STN storage. We measured the storage of SOC and STN and the content of oxidizable minerals in soils from differen...

Abstract Aim Ecological and anthropogenic factors shift the abundances of dominant and rare tree species within local forest communities, thus affecting species composition and ecosystem functioning. To inform forest and conservation management it is important to understand the drivers of dominance and rarity in local tree communities. We answer the following research questions: (1) What are the patterns of dominance and rarity in tree communities? (2) Which ecological and anthropogenic factors predict these patterns? And (3) what is the extinction risk of locally dominant and rare tree specie...
